  • Tapinarof: Dermavant Sciences

Tapinarof is a therapeutic aryl hydrocarbon receptor modulating agent (TAMA) that binds and activates the aryl hydrocarbon receptor in multiple cell types, including cells of the target tissue-human skin. Tapinarof has also shown to moderate proinflammatory cytokine expression in stimulated peripheral blood CD4+ T cells and ex vivo human skin, and impact barrier gene expression in primary human keratinocytes. The drug is currently being evaluated in Phase III stage of development for the treatment of patients with Atopic dermatitis.

Etrasimod is an oral, once daily, selective sphingosine-1-phosphate (S1P) receptor modulator designed for optimized pharmacology and engagement of S1P receptors 1, 4, and 5. Lirentelimab (AK002) is an afucosylated, humanized IgG1 monoclonal antibody which activates the inhibitory receptor Siglec-8. Additionally, Lirentelimab depletes eosinophils via antibody dependent cellular cytotoxicity (ADCC) in blood. Siglec-8 is a member of the family of cell surface receptors called Sialic acid-binding immunoglobulin-type lectins (Siglecs).
